This dataset contains genomic sequence data associated with a metabolically engineered Pichia pastoris strain developed for the heterologous production of δ-(L-α-aminoadipyl)-L-cysteinyl-D-valine (ACV), the tripeptide precursor of β-lactam antibiotics. The strain was engineered through heterologous expression of codon-optimized pcbAB (ACV synthetase) and npgA (phosphopantetheinyl transferase) genes required for activation of the non-ribosomal peptide synthetase involved in ACV biosynthesis. Adaptive laboratory evolution (ALE) was performed under glycerol-based growth conditions to improve strain performance. Whole-genome sequencing was carried out to compare the parental unevolved strain and the evolved strain isolated after prolonged cultivation. The sequences enable identification of genomic variations associated with improved cellular fitness and ACV production phenotype. The dataset includes genome assemblies of both the engineered parental strain and the evolved strain, as well as BLAST annotation results confirming the presence and genomic context of the heterologous pcbAB and npgA genes. These data support comparative genomic analysis of evolutionary adaptations occurring during strain optimization.
