Transcriptomic Profile of Neuroinflammation in Alzheimer’s Mouse Brain Treated With and Without Extracellular Vesicles

Expression of 757 genes related to neuroinflammation was analyzed across 4 treatment groups of male mice to investigate the neuroprotective effects of either human neural stem cell (hNSC) or induced pluripotent stem cell microglial-like (iMGL) cell-derived extracellular vesicles (EVs) in 5xFAD mouse model of Alzheimer’s Disease (AD). Samples from 18 male mice, ages 25-26 weeks at the time of perfusion, were analyzed using the commercially available nCounter® Mouse Neuroinflammation Panel from NanoString™, with 4-5 samples for each of the 4 treatment groups. The 5xFAD transgenic mouse line was selected for the rapid neuropathological progression of beta-amyloid plaque deposition and subsequent cognitive dysfunction. 5xFAD mice were randomly assigned to one of three AD treatment groups that would receive 4 weekly retroorbital injections of either: hNSC-derived EVs or iMGL-derived EVs at a dose concentration of 2.25E+07 EVs/50 ul injection; or vehicle consisting of phosphate buffered saline without calcium or magnesium (PBS -/-). C57BL/6J mice served as WT controls and also received 4 weekly retroorbital injections of vehicle.
