Single-cell RNA sequencing of immune cells of mouse spinal cord using Drop-Seq Overall design: Drop-Seq was performed for spinal cord immnue cells dissociated from ambisextrous C57BL/6J mice after spinal cord injury at 0, 3, and 14 day-post injury (dpi). Single-cell suspensions derived from heathy spinal cords were generated using the ActD-based protocol or standard dissociation protocol. Single-cell suspensions dissociated from injured spinal cords were generated only using the ActD-based protocol. Drop-Seq was used to produce single-cell cDNA libraries attached to microbeads. cDNAs were amplified by PCR and prepared for sequencing.
