Aneurismal subarachnoid hemorrhage (aSAH) is a neurovascular disease characterized by blood released into the subarachnoid space due to cerebral arteries rupture. After the onset of bleeding, secondary associated vasospasm (VSP) remains a dramatic side effect that causes severe comorbidities. In brain arteries from SAH (and control) animals we performed a total RNA extraction and a microarray analysis of cerebral arteries from animals 7 days after surgery to study the long-term transcriptional effects induced by SAH Male wistar rats (400 g approximately and six months old) were divided into three groups: control, sham7d, and SAH7d. 100 µL of cerebrospinal fluid was removed from the cisterna magna through the posterior atlanto-occipital membrane. Then, in the SAH group, 100 µL of blood was obtained from the caudal vein and immediately injected intracisternally. In the sham group the same procedures were performed, using the same volume of saline. The control group did not receive any surgery. For RNA analysis, whole brains were obtained and arteries were cleaned under a magnifying glass in ice-cold RNAses-free Hank’s solution, immediately frozen in liquid nitrogen and stored at -80 ºC.The homogenization of the arteries was performed the RNeasy® Micro Kit.The amplified cDNA was quantified, fragmented and labeled for hybridization with GeneChip® Clariom S Rat Array.
