The project aims to investigate transcriptional changes induced by the KRAS G12D mutation in pancreatic cancer. It involves constructing a KRAS wild-type pancreatic cancer organoid. By infecting the wild-type human pancreatic cancer organoid with KRAS-G12D lentivirus, a human pancreatic cancer organoid expressing KRAS-G12D was established. Transcriptomic analysis was performed on KRAS wild-type and G12D mutant organoids, with three samples in each group, successfully identifying transcriptional changes induced by the G12D mutation.
