Positional mapping of essential genes for growth of L. pneumophila in nutrient rich media identified several large genomic islands devoid of these genes. Targeted deletion of 6 of these islands simultaneously to generate a minimal genome strain resulted in the accumulation of mutations that partially rescued the growth defect attributed to lack of one of these islands in one isolate. Sequencing of the whole genome of two individual isolates, one partially rescued for growth (Hextuple_2q) and one that was not (Hextuple_3a) allowed the mutations responsible to be identified.
