Quantitative EBF1 ChIP-seq analysis in EBF1wt- or EBF1E271A-expressing pro-B cells harboring doxycycline-inducible NICD1 expression after doxycycline treatment. Overall design: Ebf1-/- c-kit+ lymphocyte progenitors isolated from fetal liver were first transduced with a doxycycline-inducible NICD1 expression system, selected for retroviral expression and then transduced with EBF1wt- or EBF1E271A-expressing retroviruses. Cells were sorted for retroviral expression, in vitro differentiated into pro-B cells and treated with doxycyline for 6 hours prior to EBF1 ChIP-seq analysis. Human B cell spike-in chromatin was used for spike-in normalization.
