The experiments were conducted to check whether mismatches and wobble base pairs in the upper stem of pri-miRNAs affects the DROSHA cleavage and thereby miRNA biogenesis. HCT116 cells were transfected with pCDNA3 empty, pCDNA3 wild-type or mutant pri-miRNA plasmids. Small RNA sequencing libraries of the transfected HCT116 cells were generated using the TruSeq Small RNA Library Prep kit.
