Single cell and spatial transcriptomic profile of whole CLAD and donor control lung tissue

Chronic lung allograft dysfunction (CLAD) is the leading cause of death in lung transplant recipients. CLAD is characterized clinically by a persistent decline in pulmonary function and histologically by the development of airway centered fibrosis known as bronchiolitis obliterans. We performed single-cell sequencing and spatial transcriptomic analysis of explanted tissues from human lung recipients with CLAD and donor control lungs. For the single cell dataset, whole lung tissue segments from four CLAD samples and three donor control samples were used. For the spatial transcriptomic dataset, tissue blocks from three CLAD samples and one donor control sample were used. Donor control tissue was obtained from donor lungs used for transplant that were harvested, perserved, implanted, and then trimmed to fit the recipient chest cavity with excess donor trimming preserved for research. Donor control lung tissue underwent formal pahtology evaluation that confirmed the absence of any significant lung disease.
