We aim to address the reviewer’s concerns regarding the novelty of our dataset and the lack of detailed data quality assessment and analysis. During spermatocyte development, a significant amount of cytoplasmic material is eliminated. As a result, mature and functional sperm cells are expected to contain substantially less RNA compared to somatic cells. Consequently, standard RNA-seq quality metrics, typically applied to somatic tissues, are inappropriate for sperm transcriptome analysis. This critical aspect has been overlooked in previous studies, and existing datasets predominantly represent samples with abundant RNA, likely from immature or defective sperm cells. Our dataset uniquely encompasses both samples meeting and failing conventional RNA-seq quality standards, providing a comprehensive view of sperm RNA content. This enables us to investigate the role of RNA in sperm generation and to develop novel RNA quality metrics tailored to sperm analysis. Furthermore, our data will facilitate the identification and characterization of residual RNA in mature sperm cells, and its potential impact on offspring phenotypes.
