We have previously shown that high concentrations of ATML1 specify giant cell identity whereas low concentrations do not; therefore, we asked whether high concentrations of ATML1 induce different downstream genes than low ATML1 concentrations. To identify the target genes of ATML1 at higher concentrations versus lower concentrations, we induced ATML1 to different levels in Arabidopsis inflorescence tissue and performed RNA-seq. Specifically, we treated the inducible ATML1 line, RPS5A>>ATML1, with 0.1 microM, 1 microM, and 10 microM of the inducer estradiol, and harvested tissue at 8, 16, 24, and 32 hours after induction.
