It has been recently discovered that chronic intestinal inflammation induces the expression of gankyrin, an oncoprotein which promotes development of colitis-associated cancer. To further investigate the role of gankyrin in inflammatory bowel disease, we prepared two groups of colitis mice for this study; Villin-Cre;Gankyrinf/f mice (i.e. small intestine and large intestine specific gankyrin deficient mice) and Gankyrinf/f control mice. These mice were separated into DSS treated (i.e. colitis induced) group and DSS untreated group. Surprisingly, colitis induced Villin-Cre;Gankyrinf/f mice compared to colitis induced Gankyrinf/f control mice have shown higher degree of inflammation. To understand the role of intestinal microbiota in the above phenomenon, bacterial 16S rRNA amplicon sequencing was conducted on cecum and rectum samples from the above mice groups. In addition, bacterial 16S rRNA amplicon sequencing was conducted on rectum samples of Villin-Cre;Gankyrinf/f mice and Gankyrinf/f control mice which were cohoused.
