Whole exome sequencing of transgenic mouse plasma cells

The project aimed to generate transgenic mouse models for the two major human multiple myeloma (MM) subgroups. The models are based on the specific overexpression of MMSET or Ccnd1 and Ikk2ca in mouse germinal center B cells and plasma cells using conditional gene targeting. Transgenic mice develop a plasma cell disorder with age, accompanied with MM-associated pathologies. Here, transgenic plasma cells were isolated from the spleen a/o bone marrow of diseased animals and subjected to whole exome sequencing to determine additionally acquired mutations and copy number alterations.
