【目的】对兜兰炭疽病病原菌进行鉴定,并进行室内防治药剂筛选,为兜兰炭疽病的大田防控提供参考依据。【方法】采集具有典型症状的兜兰炭疽病标本,以组织分离法进行病原物分离培养,通过形态学观察和rDNA-ITS序列分析对兜兰炭疽病病原菌进行鉴定;通过室内温度、光照控制和培养基中不同pH、碳源、氮源的差异试验确定病原菌菌丝生长、产孢量和孢子萌发条件等生物学特性;选用12种杀菌剂进行兜兰炭疽病病原菌室内药效试验,并对筛选出效果理想的药剂进行室内毒力测定。【结果】通过形态学观察和rDNA-ITS序列分析,确定分离获得的兜兰炭疽病病原菌的无性态为半知菌类黑盘孢目刺盘孢属胶孢炭疽菌(Colletotrichum gloeosporioides)。
【Objective】To identify the pathogenic fungus causing Paphiopedilum anthracnose and screen indoor fungicides, so as to provide scientific references for the field control of Paphiopedilum anthracnose. 【Method】Specimens of Paphiopedilum anthracnose with typical symptoms were collected, and the pathogen was isolated and cultured using the tissue isolation method. The pathogen was identified through morphological observation and rDNA-ITS sequence analysis. Biological characteristics of the pathogen, including mycelial growth, spore yield and spore germination conditions, were determined via experiments involving indoor temperature and light control, as well as different pH values, carbon sources and nitrogen sources in the culture medium. Twelve fungicides were selected for indoor efficacy tests against the Paphiopedilum anthracnose pathogen, and indoor virulence assays were conducted on the screened effective fungicides with optimal performance. 【Result】Through morphological observation and rDNA-ITS sequence analysis, the asexual morph of the isolated Paphiopedilum anthracnose pathogen was identified as Colletotrichum gloeosporioides, which belongs to Colletotrichum, Melanconiales, Deuteromycetes.
