We have identified a novel antimalarial compound, (S)-SW228703. This compound is relatively fast-acting, and is active against asexual blood stage parasites, liver stage parasites, and possibly gametocytes (the transmissible form). To identify the mechanism of action and/or resistance mechanisms for (S)-SW228703, we generated resistance in cultured P. falciparum parasites. Most notably, we identified mutations in the cyclic amine resistance locus (PfCARL; PfDd2_030027000) and the acetyl coA transporter (PfACT; PfDd2_100041800).
