Identification of a host factor essential for coronavirus replication Overall design: A CRISPR/Cas9 library was used to identify genes that rendered the cell refractory to killing by hCoV-229E. The complete sgRNA librarywas packaged into lentivirus and used to transduce Huh7 cells . In two independent biological replicates, the transduced cells were infected with the virus, and the cells that were not killed by infection were collected and the sgRNAs enriched in those populations were sequenced.
