Dietary intake of whole grains and legumes have been reported to reduce colorectal cancer (CRC) risk. A pilot, randomized, placebo-controlled, two arm trial was implemented to evaluate dietary intake of rice bran and navy beans for 12 weeks while accounting for physical activity levels (n = 20). Adults with polyps or stage 1 cancer removed in the last 3 years were eligible and randomization was by body mass index, sex. Participants assigned to the intervention consumed 30g heat-stabilized rice bran + 30g of cooked navy beans powder daily. Sixteen study foods were prepared with and without rice bran + navy bean powder. Fibersol -2 (10g/ day) was the placebo. Non-targeted and targeted metabolite profiling was performed on blood, urine, and stool before, during, and after dietary intervention to measure changes in metabolic pathways associated with reduce colonic inflammation and cancer progression. Stool was analyzed for primary and secondary bile acids, short chain fatty acids and 16S gene sequencing, while stool, plasma and urine were subjected to non-targeted metabolomics by UPLC-MS/MS.
