Single-cell RNA sequencing was employed to investigate the host-virus interaction between Mandarin fish (Siniperca chuatsi) and Infectious spleen and kidney necrosis virus (ISKNV). Comparative analysis was performed on spleen samples obtained from ISKNV-infected individuals and healthy controls. Overall design: Comparative single-cell RNA sequencing was performed to investigate the splenic immune response of Siniperca chuatsi to ISKNV infection. The study design consists of two experimental groups: an ISKNV-infected group (sampled at 7 days post-infection) and a matched mock-infected control group (PBS-injected). To minimize individual heterogeneity and ensure biological representativeness, each sequenced sample represents a pool of spleen tissues collected from six distinct biological individuals. The experiment aims to characterize cell-type-specific transcriptional changes and immune dynamics induced by the virus compared to the healthy control.
