To explore the impact of tet(X4) expression on bacterial metabolomic state, tet(X4) gene was cloned into a high-copy plasmid, pUC19, to generate pUC19-tet(X4). An empty vector without tet(X4) (pUC19) was included as the control. Subsequently, tet(X4) and pUC19 plasmid was transformed into reference strains E. coli DH5α. Then, we performed metabolomic profiling of tet(X4)-negative and -positive bacteria using Gas Chromatography-Mass Spectrometry (GC-MS).
