Polycations for gene delivery: deepening their behaviour at cell surface

Polycations can efficiently condense plasmid DNA (pDNA) to safely reach target cells without activating the immune system. However, the role of polycations structure on the interaction with cells membranes and toxicity of polyplexes remain unclear. The aim of our work is to develop and optimize innovative formluations of polycations with different architectures (linear and 3-4 arms star), condensated with DNA. To ptimixe their efficiency with cell-free strategies, we aim at understanding the molecular details of different nanovectors interaction with complex biomimetic membranes. Not only the interaction at cell surface will be mimed, but also the interaction once in endosomal environment.
