This dataset supports a research project investigating age-associated changes in cardiac lysosomal organisation using a combination of IVIS Lysotracker imaging, qPCR-based gene expression profiling, and R-based statistical modelling. The study aimed to determine whether ageing alters the abundance of acidic vesicles or the acidification machinery in murine hearts.Whole-heart acidic vesicle signals were quantified using IVIS two-dimensional (2D) fluorescence imaging from both anterior and posterior orientations, enabling rapid acquisition (~10 seconds per heart). RT-qPCR analyses were performed for lysosomal and autophagy-related markers, including Lamp2, Atp6v1a, Sqstm1, Cd63, and M6pr, to evaluate transcriptional remodelling. R scripts were used to perform linear mixed-effects modelling to assess the effects of age, region, and batch on imaging and qPCR results.The repository includes raw fluorescence intensity data, qPCR cycle threshold (Ct) values, and all scripts used for statistical analysis. These data support the reproducibility of the study and provide a resource for researchers interested in cardiac ageing, lysosomal biology, vesicular trafficking, and imaging-based quantification of acidic compartments.
