Gain-of-function mutations in PI3K result in a human primary immunodeficiency syndrome characterized by lymphopenia, lymphoproliferation, recurrent respiratory infections and inefficient responses to vaccination. However, what promotes this constellation of immune disturbances at the cellular and molecular level remains unknown. A mouse model that recapitulates major features of this disease was generated and the microbiota of these mice compared to wildtype was characterized using 16s rRNA sequencing. Additionally, IgA repertoires against fecal bacteria were profiled by sorting of IgA-coated fecal bacteria from IgA-uncoated bacteria and 16s rRNA sequencing both pools. PI: Yasmine Belkaid (NIAID).
