Gene expression profiling of purified stem and progenitor cell types from femoral fracture calluses of patients hemiarthroplasty or open reduction internal fixation procedures. We used microarrays to detail the global program of gene expression of stem and progenitor cells responsible for skeletal repair. Human skeletal stem and progenitor cells were purified from femoral fracture tissues via flow cytometry for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain homogeneous populations of hSSCs, hOPs and hCPs in order to compare the cell type specific expression profiles. To that end, we isolated CD45-CD235a-CD31-TIE2-PDPN+CD146-CD164+CD73+ hSSCs, CD45-CD235a-CD31-TIE2-PDPN-CD146+ hOPs and CD45-CD235a-CD31-TIE2-PDPN+CD146-CD164-/+CD73-/+ hCPs.
