In this study, we developed a drug delivery nanoplatform for triple-negative breast cancer (TNBC) treatment by using human red blood cell-derived extracellular vesicles (RBC-EVs) loaded in the lumen with Cisplatin and functionalized on the membrane with Cetuximab (CTX) for EGFR targeting. We investigated the molecular mechanisms underlying the cytotoxic effects of different Cisplatin-loaded RBC-EV formulations by performing the RNA-seq transcriptome profiling of MDA-MB-231 cells after 24 hours of different treatments as well as untreated cells as control. Overall, our results demonstrated the promising potential of Cisplatin-loaded CTX-functionalized EVs for efficient and selective combined therapy of chemoresistant TNBC.
