Analysis-ready AlamarBlue, LDH, and RT-qPCR dataset for an in vitro MDCK low-MOI LPAIV xenobiotic exposure study

This dataset contains the analysis-ready data used for the study entitled “Environmentally relevant xenobiotics decouple influenza viral burden from infected-cell metabolic phenotype in a readout-optimized in vitro model ”. The dataset was generated from an in vitro MDCK cell infection model designed to evaluate how an environmentally informed xenobiotic mixture modifies low pathogenic avian influenza virus (LPAIV)–host cell interactions. MDCK cells were exposed to a xenobiotic mixture containing TCDD, PCB126, and 9-ClPhe and infected at low multiplicity of infection (MOI 0.01) with one of four LPAIV subtype backgrounds: H16N3 (V1), H5N3 (V2), H7N3 (V3), or H9N2 (V4). Measurements were collected at 6, 12, 24, and 48 h post-infection across four experimental repeats. The file includes experimental metadata and matched assay measurements for AlamarBlue metabolic activity, LDH release, and RT-qPCR-based extracellular viral RNA quantification. AlamarBlue readouts include fluorescence and absorbance measurements at 1, 2, 3, and 4 h incubation windows, enabling empirical comparison of readout mode and incubation duration. LDH measurements are provided as OD490 values. RT-qPCR values are provided as mean Ct values calculated from duplicate PCR reactions, where lower Ct values indicate higher extracellular viral RNA abundance. Technical well pairs were averaged before inclusion in this analysis-ready dataset. The dataset also contains condition annotations, virus identifiers, xenobiotic/vehicle status, infection status, blank and 100% reduced AlamarBlue reference groups, LDH normalisation context, qPCR applicability, source well positions, and technical-pair quality-control fields. These data support analyses of AlamarBlue readout optimisation, time-dependent extracellular viral RNA accumulation, virus- and time-dependent xenobiotic-associated changes in extracellular viral RNA, infected-cell metabolic activity, LDH release, and integrated Ct–phenotype relationships.
