We generated Drosophila melanogaster lines with the insertion of 16 endogenous boundaries in 2 orientations into 2 acceptor TADs (TAD A scyl-chrb, TAD B sna-esg). To inspect local changes in chromatin topology, we performed tiled capture Hi-C in 4-20 hour embryos from wild type and insertion lines with probes capturing the 2 acceptor TADs and 3 background regions.
