Despite significant therapeutic potential of SWI/SNF inhibition therapy, the off-tumor effects of SWI/SNF inhibition remain largely unknown. Here, we quantify the effects of the dual SWI/SNF ATPase inhibitor BRM014 on peripheral blood (PB) and bone marrow (BM) cells in mice. We identify altered chromatin regulatory mechanisms in B cells and monocytes. Overall design: Single-nuclei joint RNA-seq (snRNA-seq) and ATAC-seq (snATAC-seq) of peripheral blood (PB) or bone marrow (BM) cells from C57BL/6 mice that received 14-day treatment of daily 20 mg/kg BRM014 or vehicle control via oral gavage. All samples are pooled from 3 independent mice in the same condition and processed using 10x Multiome ATAC + Gene Expression kit according to the manufacturer's instructions.
