This project investigates the transcriptional network regulated by GhMYB44 and its downstream factor GhFSN43 during secondary cell wall (SCW) biosynthesis in cotton fiber. RNA-seq was performed on 20 days post-anthesis (DPA) fiber samples from GhMYB44 overexpression (OE), RNA interference (RNAi), and GhFSN43 overexpression (OE) and knockout (KO) lines, using Gossypium hirsutum cv. CRI24 as the wild-type control. In addition, DAP-seq reads of GhMYB44 were generated to identify its genome-wide DNA binding sites. These datasets together provide a comprehensive resource for understanding the molecular mechanisms underlying transcriptional regulation of SCW biosynthesis in cotton fibers.
