Oxygen/glucose deprivation and reoxygenation induces oxidative stress in BMDMs

To gain additional insight into the potential role of oxidative stress at the mitochondria, we performed RNA seq of BMDMs subject to oxygen/glucose deprivation and reoxygenation (OGD/R). BMDMs from male and female mice were differentiated by L929 supernatant.To simulate ischemia reperfusion injury, a glucose oxygen deprivation and reoxygenation model was used. Cells were plated in glucose free 1640 RPMI medium and placed in a three gas incubator for 6 hours of hypoxia treatment. After that, 10% FBS and 2g/L glucose were added to the medium followed byincubation under normal oxygen conditions for 1 hour before harvest. The control group (NC) was cultured in a complete medium of 1640 RPMI supplemented with 10% FBS. Total RNA from adherent BMDMs was extracted using the Triol method, and second-generation transcriptome sequencing was performed.
