Objective: The functional significance of PDE1B in CRC cell lines was investigated through flow cytometry.Methods: The Annexin V-FITC Apoptosis Detection Kit (E-CK-A211, Elabscience, China) was used to perform apoptosis analysis following the manufacturer's instructions. HCT116 and SW480 cells from the different treatment groups were trypsinized, collected by centrifugation at 1,000 rpm for 5 min, and then washed with cold 1× PBS. Subsequently, the cells were incubated with propidium iodide (PI) and annexin V reagents in the dark at room temperature for 15 min. Flow cytometric analysis was performed using a BD FACSAria IIIu instrument, and apoptotic rates were quantified using FlowJo software (version 10.10, USA).Results: Flow cytometry results showed that the PDE1B-overexpressing group exhibited a significantly higher proportion of apoptotic cells compared to control cells, suggesting that PDE1B overexpression promotes apoptosis in CRC cells.
