To investigate in vivo off-target profile of xABE, we collected RNA from right xABE injected and left uninjected TA tissue for RNA-seq and SNV (single nucleotide variant) identification analysis. CBh- and MHCK7-xABE were evaluated and found to generate similar amount of SNV with control sample (TA without xABE treatment), indicating undetectable in vivo off-target effect for xABE even when therapeutic effect achieved.
