A protocol modified from Chen et al. (2019) was established to quickly and reproducible induce a large set of genes after short-term heat-shock induction of A.thaliana seedlings at 44 degree Celsius followed by extraction of nuclei, RNA isolation and Next-Generation Sequencing in several transcription elongation factor knockout mutant lines.With this method we could not only see differential expressed genes within the lines after heat shock induction but also the distribution of reads within the genes which revealed specific stop points of transcription.
