The first draft of the T. cruzi genome, assembled using Sanger sequencing, revealed a highly repetitive structure, with a significant portion of these repeats corresponding to genes encoding surface proteins. Advances in sequencing technologies, such as Nanopore and PacBio, have enabled the generation of high-quality assemblies. However, the latest PacBio Sequel Hi-Fi technology has, for the first time, made it possible to assemble the diploid T. cruzi genome into 32 complete chromosomes, from telomere to telomere.
