PROBLEM TO BE SOLVED: To provide alternative and robust systems and techniques for versatile sequence targeting. SOLUTION: Provided is a non-naturally occurring or engineered composition comprising (A) a first regulatory element operably linked to a CRISPR-Cas based chimeric RNA polynucleotide sequence and a second regulatory element operably linked to an enzyme coding sequence encoding a CRISPR enzyme comprising at least one or more nuclear localization sequences, or (B) a guide sequence that can hybridize to a target sequence in an eukaryotic cell, and a first regulatory element operably linked to at least one or more tracr mate sequences, a second regulatory element operably linked to the sequence encoding the CRISPR enzyme, and a third regulatory element operably linked to a tracr sequence, each element being on the same or a different vector, the CRISPR enzyme being derived from a microorganism of a particular genus.
