Bulk RNAseq of fetal human lung fibroblasts (MRC5) and primary adult human lung mesenchyme (AHLM)

The fetal human lung fibroblasts (MRC5) and primary adult human lung mesenchyme (AHLM) have different capabilities in supporting human alveolar type 2 cell transdifferentiation into basal KRT5+ cells.Therefore, we examined the transcriptome of AHLM vs. MRC5 by bulk RNAseq. MRC5 and AHLM cells were cultured in DMEM/F12 with 10% FBS for 2 days. Then the total RNA were extracted from technical triplicates of MRC5 and AHLM cells, respectively. The RNA of the samples were sent to GENEWIZ company for sequencing. Raw sequence data (.bcl files) generated from Illumina HiSeq was converted into fastq files and de-multiplexed using Illumina's bcl2fastq 2.17 software.
