Alveolar epithelial type 2 (AT2) cells are facultative progenitor cells that drive adult alveolar regeneration after acute lung injury. Using transcriptomic analyses from in vivo mouse injury models, we define the role of Tfcp2l1 in regulating AT2 cell behavior during lung regeneration. To determine the role of Tfcp2l1 in AT2 cells, we generated SftpcCreERT2; R26REYFP or SftpcCreERT2; Tfcp2l1fl/fl; R26REYFP mice, allowing us to inactivate Tfcp2l1 in AT2 cells and trace their fate. To assess whether Tfcp2l1 regulates AT2 cell-mediated lung regeneration, each mouse line received three doses of 200mg/kg tamoxifen and 14 days after induction; these mice were infected intranasally with 50μL of a 1LD50 dose of mouse-adapted H1N1 PR8 influenza virus. 14 days post-viral infection, mouse lungs were isolated, and lineage traced cells (YFP+) were cell sorted to generate respective libraries for genome profiling
