The miRNAs associated with immunotoxicity to gossypol in fish have not been fully explored.Constructing a miRNA expression profile of fish spleen cells exposed to gossypol could enhance our comprehension of the role of miRNAs in modulating the functions of the immune systems. This research aimed to identify the specific miRNAs responsible for the immunotoxicity of gossypol on the spleen of Cyprinus carpio. The fish were assigned to gossypol group (oral administration 0.1 mL 20 mg gossypol acetate) and control group (oral administration 0.1 mL normal saline) at random, fish received a continuous oral administration of normal saline or gossypol acetate for a week. 24 h after the seventh oral administration, fish were randomly selected in each group, then the fish were anesthetized with MS 222 , 10 spleens (3 or 4 fish in each tank) were stored for miRNA sequencing. Six miRNA libraries: gossypol 1, gossypol 2, gossypol 3, control 1, control 2, and control 3 were constructed and sequencing.
