The imbalance of osteogenesis and adipogenesis is one of the major cause of osteoporosis. Mesenchymal stem cells (MSCs), with the potential differentiated into osteocytes and adipocytes, provide useful tools to investigate osteoporosis. Circular RNAs (circRNAs) play crucial roles in multiple biological processes; however, their characteristic properties during the bilineage differentiation of MSCs remain poorly described. Here, we established osteogenesis and adipogenesis system using human MSCs derived from bone marrow and adipose, and performed the identification by flow cytometric, histological and quantitative PCR analysis. Then we generated a rich data set of high-throughput RNA sequencing of human MSCs during bilineage differentiation at five time points. Our data is consisted of 50 libraries from 30 samples with the RiboMinus, RiboMinus/RNase R or Poly A enrichment treatment, each library obtaining an average sequencing depth of 62 million reads. Several tens of thousands circRNAs were identified from the integrated analysis of RNA treated with different method, among which several hundreds of lineage-specific circRNAs provide the potential regulatory components for the imbalance of osteogenesis and adipogenesis of MSCs.
