To approximate telomere crisis in vitro, Titia de Lange's lab generated a derivative of the hTERT ex- pressing RPE-1 retinal pigment epithelial cell line in which theRband p53 pathways were disabled with shRNAs to Rb and p21. To induce the telomere fusions typical of telomere crisis, they used a dox-inducible dominant negative allele of TRF2 (TRF2-DN), which deprotects telomeres and induces telomere fusions. This was then used to generate populations of cells that had experienced telomere crisis, which were then expanded as single-cell derived clones for genomic analysis. Here, we sequence at low coverage a large number of cells to identify those with evidence of copy number changes indicative of telomere crisis. Those with telomere crisis will then be sequenced at higher depth to identify patterns of structural variation and point mutations.
