Since the gut microbiota-brain axis is an emerging area of research and has great potential for therapeutic applications, we investigated the effect of microbial remodeling on theta oscillation and spatial memory in a 5xFAD AD model mice using a prebiotic approach. We have determined that feeding 5xFAD mice a mixture of non-digestible saccharides, RMD-101, resulted in a reduction of elevated theta power during REM sleep, reduced levels of serum tau and improvement in hippocampal-dependent spatial memory consolidation. We also found that RMD-101 increased the level of hippocampal acetylcholine (Ach), a regulator of theta activity in the hippocampus. Feeding study mice RMD-101 selectively enriched gut bacteria genus, Parabacteroides, that carry the choline-phosphate cytidylyltransferase enzyme, which we have shown to catalyze CDP-choline synthesis in vitro.
