To identify the genome-wide binding sites of lactylated NPM1 and understand its role in transcriptional regulation. Overall design: We generated NPM1-knockout (NKO) HK-2 cells followed by reconstitution with WT NPM1 (NKO-NPM1 WT) or the K257R mutant (NKO-NPM1 K257R).Fragmented chromatin was immunoprecipitated overnight at 4°C with anti-NPM1 antibody or rabbit IgG, followed by incubation with protein G-agarose beads.
