By RNA-sequencing, we compared WT vs ST2 knockout leukemia stem cells (LSCs) from the bone marrow of ST2flox/flox and ST2flox/flox Mx1Cre MLL-AF9 LSCs, and found that ST2 deficiency in LSCs reprogramme the oncogenic, metabolic and cell cycle signatures which correlated with reduced leukemogenesis and maintenance. Overall design: ST2flox/flox LSCs (control) and ST2flox/flox Mx1Cre LSCs were injected into lethally irradiated C57BL/6 mice at Day 0, and polyinosinic:polycytidylic acid (pIpC) was administrated every other days for a total of 7-doses started on the same day. BM cells were obtained and two types of LSCs following fluorescence-activated cell sorting were deliveried for RNA extraction, library construction, and RNA-sequencing.
