Aged mice (22-24 mo) underwent PoWeR for 8 weeks. RRBS on myonuclei from soleus muscles (n=3 sedentary and n=3 trained) were subjected to low-input RRBS. Overall design: Human skeletal actin promoter reverse tetracycline transactivator tetracycline response element driven histone 2B green fluorescent protein mice (termed HSA-GFP) were generated to label resident myonuclei at the onset of experimentation. Myonuclera were isolated via fluorescent activated myonuclear sorting, then DNA was isolated from purified myonuclear using the Qiagen DNA Micro Kit using carrier RNA. Low-input reduced representation bisulfite sequencing was carried out by Zymo Research.
