The study was designed to investigate the acute effects of B2AR receptor activation on global gene expression changes in bone marrow-derived macrophages. Fenoterol was chosen as B2AR agonist, but our earlier experiments using B2AR antagonists and B2AR knockout cells indicated that norepinephrine signal predominantly via B2AR in macrophages. Bone marrow cells were isolated from 4 mice and differentiated to macrophages in vitro. Cells from each mouse were either untreated (control) or treated with beta2-adrenergic receptor agonist fenoterol for 1 or 4 hours, in the presence of BMDM culture medium. Cells were then lysed and total RNA was isolated for sequencing.
