The aim of this experiment was to analyze differences in gene expression between synovial myeloid (CD11b+Gr-1+) cells from wild type (C57BL6; WT, B6) and Fas-/- mice with antigen-induced arthritis (AIA). Immunized control (CTRL) mice were used to asses the change in synovial myeloid cell trascriptome after arthritis induction in both genotypes. AIA was induced in wild-type (WT, B6) and Fas-/- mice by immunization with methylated bovine serum albumin (mBSA) and subsequent intra-articular injection of mBSA. Immunized control (CTRL) mice were intra-articulary injected with phosphate buffered saline (PBS). Ten days after intra-articular injection knee joints were harvested and synovial cells were released by collagenase and labeled with anti-mouse CD45-FITC, CD11b-PE, Gr1-PECy7, B220/CD3/NK1.1/CD31/TER119-APC, and CD51-APCeF780. CD11b+Gr-1+ population was sorted using BD FACSAria, RNA extracted by Trizol, and hybridized to Affymetrix Mouse Gene ST 2.0 arrays. Each experimental group contained 4 samples.
