We report that MAdCAM and retinoic acid (RA), two constituents of gut tissues, together with TGF-b, promote the differentiation of CD4+ T cells into a distinct subset a4b7+ CD69+ CD103+ TRMs. These findings provide a framework to better understand the contribution of CD4+ TRMs to persistent viral reservoirs and HIV pathogenesis. CD4+ T cells from 6 healthy donors were subjected to 8 different treatments with 5 reagents (i.e. anti-CD3 Ab, anti-CD30 Ab, cV2, MadCAM, RA). The mRNA was extracted from approximately 2 million human T-cells for each sample, and subjected to 75-bp RNA-Seq sequencing by Illumina NextSeq.
