We report that PRRX2 mediates enzalutamide resistance by activating the anti-apoptotic BCL2 and theE2F/RB1 cell cycle pathways. We used LNCaP cells to over-express the transcription factor PRRX2 using CRISPR activation. Negative Control (LNCaP-sgNC) and PRRX2 over-expressing cells (LNCaP-sgPRRX2) were treated with the AR inhibitor enzalutamide (5uM) for 4 days. We then studied gene expression changes in both cell lines treated with enzalutamide or DMSO.
