The cellular mode of actions of inhaled nanomaterials are poorly understood, though shape and biopersistence of nanomaterials long-since raise concerns in respiratory toxicology. Here we longitudinally analyzed cellular perturbations to three carbonaceous nanomaterials of different shape and toxicity upon pulmonary delivery to mice at single cell resolution. Focusing on the course of lung inflammation, our research aims to reveal highly nanomaterial-specific contributions of cell niches. Overall design: Four 10-12 week old, female C57BL/6 mice were anesthetized with midazolam/ medetomidine/fentanyl (MMF) and instilled intratracheally with 50 µl of either carbon nanoparticle (CNP; 20 µg), multi walled carbon nanotube (MWCNT; 15 µg) or double walled carbon nanotube (DWCNT; 50 µg), 0.1 µg LPS or sham (0.5 mg/mL lung surfactant in water) per mouse. Whole lung tissue was harvested from 4 mice per group after 12h, 6d or 28d for live single cell and tissue analyses (Drop Seq).
