A caffeine-resistant Saccharomyces cerevisiae mutant strain was obtained using an evolutionary engineering strategy based on successive batch cultivation at gradually increasing caffeine levels. The mutant strain Caf905-2 was selected at a caffeine concentration where its reference strain could not grow at all. Whole-genome transcriptomic analysis of Caf905-2 was performed with respect to its reference strain. Agilent Yeast One-Color DNA microarray systems were employed for comparative whole-genome transcriptomic analysis of the selected mutant Caf905-2 and its reference strain. Before gene expression analysis, each culture was grown in triplicate in yeast minimal medium (YMM) at 30°C and 150 rpm. Total RNA was isolated, using RNeasy Mini Kit (Qiagen) at early exponential phase of growth (OD600 ~1.0).
