Prokaryotic communities in various biofilter carriers from Japanese drinking water treatment plants (DWTPs) were investigated using 16S rRNA gene (16S rDNA) amplicon sequencing. Samples included sand from slow sand filters (SA) and biological activated carbon (BAC) from ozone-BAC filters (BACFs) and up-flow biological contact filters (U-BCFs). Both DNA extracts and cDNA PCR products (reverse transcribed from RNA) were sequenced to capture total and potentially active microbial communities, respectively. The study aimed to compare microbial communities across different carriers and explore the impact of factors such as location, biological filter type, nucleic acid type (DNA vs. RNA), and seasonal variation. Sample nomenclature includes abbreviations for filter type (SA, BACF, UBCF) and DWTP location (TK, CB, HG, OS, FO), followed by a number (if available) indicating different biological filters and sampling time, respectively. An additional sample type, HMA, represents UBCF samples from a specific DWTP, with numbers denoting sampling month.
